ATP level and respiration of embryos.

چکیده
A considerable increase in the rate of oxygen consumption is known to occur during the development of embryos (cf. Needham, 1931; Tuft, 1953; Brachet, 1960; Gustafson, 1965). However, the mechanism of the increase in respiration during embryonic development is still unclear. A certain correlation seems to exist between DNA synthesis and the respiration of embryos (Comita & Whiteleley, 1953; Brachet, 1960). According to Commoner (1964), this correlation is determined by a change in the level of free nucleotides in cells when the rate of DNA synthesis changes. Free nucleotides control cell metabolism, oxidative processes in particular. The ADP+P/ATP ratio is known to control the rate and direction of electron transfer in the respiratory chain (Chance & Hagihara, 1961; Klingenberg & Schollmeyer, 1961). The ADP/ATP system is suggested to control the rate of oxidative metabolism in fertilization (Monroy, 1965 a, b; Zotin, Milman & Faustov, 1967). The work described below was aimed at the elucidation of the role played by ATP in respiration changes of embryos during their development and during changes in environmental temperature.

Mitochondria and human preimplantation embryo development.
Human reproduction, like all biological systems, is characterised by a large level of variability. In this field, the variability is observed as a large difference in implantation potential of human embryos developing in vitro, despite similarities in observable parameters such as rate of development and morphology of these embryos. One of the underlying factors that determines developmental po...
متن کاملThe Effect of Vitrification and in vitro Culture on the Adenosine Triphosphate Content and Mitochondrial Distribution of Mouse Pre-Implantation Embryos
Background: The mitochondria are an important source of adenosine triphosphate (ATP) production in pre-implantation embryo. Therefore, the objective of this study was to investigate the effect of vitrification and in vitro culture of mouse embryos on their mitochondrial distribution and ATP content. Methods: The embryos at 2-PN, 4-cell and blastocyst stages were collected from the oviduct of st...
متن کاملInvolvement of Cytochrome P-450 in n-Butyl Nitrite-Induced Hepatocyte Cytotoxicity
Addition of n-butyl nitrite to isolated rat hepatocytes caused an immediate glutathione depletion followed by an inhibition of mitochondrial respiration, inhi- bition of glycolysis and ATP depletion. At cytotoxic butyl nitrite concentrations, lipid peroxidation occurred before the plasma membrane was disrupted. Cytochrome P-450 inhibitors inhibited peroxynitrite formation and prev...

A Thai plant Clitoria macrophylla Wall. was claimed to possess some pharmacological activities. The phytochemical studies reported a rotenoid compound, 6-deoxyclitoriacetal, which exhibits the cytotoxic effect in several cell line experiments. This report was to study the effect of 6-deoxyclitoriacetal on rat liver mitochondria, which may associated to its cytotoxic phenomena. Mitochondrial sus...
